چکیده

New colorimetric methods are described for determination of sub-milligram amounts of ultra-high molecular weight polyethylene (UHMWPE) wear particles. These methods are based on the irreversible binding of the fluorescein-conjugated bovine serum albumin or the hydrophobic dye Oil Red O to wear particles. UHMWPE particles bind both substances from their solutions and thus decrease the absorbance of these solutions. The decrease is linearly dependent on the amount of added wear particles in the sub-milligram range suitable for practical use. The newly developed method offers improved accuracy and precision compared to Fourier transformed infrared spectroscopy (Slouf M, et al. Quantification of UHMWPE wear in periprosthetic tissues of hip arthoplasty: description of a new method based on IR and comparison with radiographic appearance. Wear 2008;265:674-684.).

Generation of a large volume of clinically relevant nanometre-sized ultra-high-molecular-weight polyethylene wear particles for cell culture studies

It has recently been shown that the wear of ultra-high-molecular-weight polyethylene in hip and knee prostheses leads to the generation of nanometre-sized particles, in addition to micron-sized particles. The biological activity of nanometre-sized ultra-high-molecular-weight polyethylene wear particles has not, however, previously been studied due to difficulties in generating sufficient volume...

Influence of third-body particles originating from bone void fillers on the wear of ultra-high-molecular-weight polyethylene

Calcium sulfate bone void fillers are increasingly being used for dead space management in infected arthroplasty revision surgery. The presence of these materials as loose beads close to the bearing surfaces of joint replacements gives the potential for them to enter the joint becoming trapped between the articulating surfaces; the resulting damage to cobalt chrome counterfaces and the subseque...

Investigation of Wear Behavior of Biopolymers for Total Knee Replacements Through Invitro Experimentation

The average life span of knee prosthesis used in Total Knee Replacement (TKR) is approximately 10 to 15 years. Literature indicates that the reasons for implant failures include wear, infection, instability, and stiffness. However, the majority of failures are due to wear and tear of the prosthesis. The most common biopolymer used in TKR  is Ultra High Molecular Weight Polyethylene (UHMWPE). Pr...

Nanomechanical and surface properties of rMSCs post-exposure to CAP treated UHMWPE wear particles

UNLABELLED Wear debris generated by ultra-high molecular weight polyethylene (UHMWPE) used in joint replacement devices has been of concern due to reductions of the implant longevity. Cold atmospheric plasma (CAP) has been used to improve the wear performance of UHMWPE. Our aim was to investigate the elastic and adhesive properties of rat mesenchymal stem cells (rMSCs), through AFM, after expos...

Identification of nanometre-sized ultra-high molecular weight polyethylene wear particles in samples retrieved in vivo.

Nanometre-sized particles of ultra-high molecular weight polyethylene have been identified in the lubricants retrieved from hip simulators. Tissue samples were taken from seven failed Charnley total hip replacements, digested using strong alkali and analysed using high-resolution field emission gun-scanning electron microscopy to determine whether nanometre-sized particles of polyethylene debri...
